Spesifik COX-2 İnhibitörü Selekoksib
Abstract
Non-steroidal anti-inflammatory drugs (NSAID’s) are effective by inhibiting cyclooxygenase enzyme responsible for prostaglandine synthesis that mediates pain and inflammation. Cyclooxygenase exists as two isoforms, cyclooxygenase-1 (COX-1) and cyclooxygenase-2 (COX-2). NSAID’s inhibit both of these two isomers. Inhibition of COX-1 is responsible for the gastric and renal toxic effects of these drugs which can cause serious damages such as cardiac insufficiency and renal function impairment, primarily gastrointestinal (GI) toxicity, in spite of their benefits.
Celecoxib as a selective COX-2 inhibitor, inhibits only COX-2 without causing toxic effects related to COX-1 inhibition by conventional NSAID’s.
